10. Estimation of Drotaverine Hydrochloride in Human Plasma

Abstract. A sensitive and specific liquid chromatography with ultraviolet detection was developed for the estimation of drotaverine hydrochloride (DH), in human plasma. Drotaverine hydrochloride and internal standard diclofenac sodium were extracted from plasma by a mixture of dichloromethane–isopropyl alcohol (80:20, v/v). The compounds were separated on a Thermo BDS Hypersil C8 (250 X 4.6 mm, 5 mm, Thermo) column. The mobile phase containing a mixture of acetonitrile and 0.02 M ammonium acetate buffer (55:45) at pH = 3.5 was pumped at a flow rate of 1 mL min-1 with UV detection at 230 nm. Recovery from plasma was 98.68%.
