Valdecoxib and Paracetamol in Tablet Dosage Form by HPLC

Abstract:
An analytical method is reported for the simultaneous
determination of valdecoxib and paracetamol in tablet dosage form by
RP-HPLC. The drugs were monitored with a diode array detector at the
fixed wavelength (245 nm). The elution was performed on a
Phenomenex C18 column pack ODS (5µ,
250 mm x 4.6 mm) and the mobile
phase consists of methanol and water (60:40; v/v) mix with the flow
rate of 1 ml.min-1. The linearity of the method ranged from
40-60 µg.mL-1 for paracetamol (r2=0.998) and
1.6-2.4 µg.mL-1 for valdecoxib (r2=0.9988) in
binary mixture. The mean recoveries were 98-101.75% for paracetamol
and 98.70-100% for valdecoxib respectively. The active constituents
were eluted at 3.17 and 7.3 min respectively for paracetamol and
valdecoxib, which are free from any interfering endogenous peaks
during a run time of 20 min. The assay was found to be highly
selective due to the absence of interfering peaks and can be
successfully applied to the determination of these active components
in pharmaceutical preparations.
